چکیده

The propagation of viruses in cultures of mammalian tissue cells provides many new applications and advantages for virus research and routine diagnostic work. Although herpes simplex virus has been propagated in tissue culture by several workers (literature reviewed by Sanders, Kiem and Lagunoff, 1953), there still exists a need for a simple method which will produce high yields of virus and give visible evidence of viral multiplication. Such a method would be valuable for the routine work of isolation, identification and titration of the virus, as well as for antibody assays. In our studies on herpes simplex virus, stationary plasma-free tissue cultures of rabbit kidney were used to propagate the virus. The results of these investigations are reported in the present paper.

Application of McCoy Cell Line for Propagation of Herpes Simplex Virus Type 1

Herpes simplex virus types 1 (HSV-1) and 2 (HSV-2) are members of the Herpesviridae family. About 40% to 80% of the world populations are infected with HSV and its prevalence is high in Iran. The high prevalence of this virus in the community and the ability of the virus in causing fatal diseases among immunocompromised patients, have encouraged studies to be performed on HSV and suitable cell ...

Replication Characteristics of Herpes Simplex Virus Type-1 (HSV-1) Recombinants in 3 Types of Tissue Cultures

A complication in the analysis of the role of ICP34.5 gene in the herpes simplex virus type-1 (HSV-1) lifecycle is the presence of overlapping antisense gene, open reading frame P (ORF P), which is also deleted in HSV-1 ICP34.5 negative mutants. A HSV-1 wild type strain (17+) ICP34.5/ORF P deletion mutant (1716) is totally avirulent in animal models and impaired in a number of in vitro function...

Detection of herpes simplex virus by 8 h in shell vial cultures with primary rabbit kidney cells.

Shell vial (SV) cultures for herpes simplex virus using primary rabbit kidney cells stained at 8 h after inoculation were compared with 20-h SV cultures as well as conventional tissue culture. Of the 326 clinical specimens examined, conventional culture detected 67, and of these, 61 (91%) and 42 (63%) were detected by 20-h SV cultures and 8-h SV cultures, respectively.

Photoinactivation of latent herpes simplex virus in rabbit kidney cells.

The photoinactivation of actively and nonactively growing herpes simplex virus by neutral red and proflavine was studied in rabbit kidney cells. Active virus growth was inhibited by both dyes under conditions which did not destroy the cells. Neutral red caused a much greater inhibition than proflavine. Neutral red also caused a reduction in the reactivation rate of latent virus when the infecte...

Protection by polyinosinic-polycytidylic acid complex of rabbit eye tissue cultures infected with herpes simplex virus. Effect of neomycin and virus challenge dose.

The induction of the interferon mechanism in rabbit corneal, conjunctival, iris, and kidney tissue cultures with polyinosinic-polycytidylic acid complex protects against infections with herpes simplex virus. The degree of protection is inversely proportional to the viral challenge dose and is significantly enhanced by the addition of neomycin. The potential application of these observations to ...
